DENVER, Colo. (247marketnews.com) — Biotechnology is delivering a fresh wave of developments that could reshape investor expectations for Disc Medicine (NASDAQ:IRON), Silence Therapeutics (NASDAQ:SLN), NeOnc Technologies Holdings (NASDAQ:NTHI), and Alaunos Therapeutics (NASDAQ:TCRT).
Disc Medicine: Phase 3 success brings bitopertin closer to regulatory milestone
Disc Medicine (NASDAQ:IRON) announced positive topline results from its pivotal Phase 3 APOLLO trial of bitopertin in patients with erythropoietic protoporphyria (EPP), including X-linked protoporphyria (XLP). The study met both co-primary endpoints and all secondary endpoints, delivering a potentially important development for patients who experience severe pain and other symptoms following sunlight exposure.
The reported results were notable across multiple measures. Bitopertin achieved a 41.4% reduction in whole-blood metal-free protoporphyrin IX (PPIX) relative to placebo, with a p-value below 0.0001. Patients receiving treatment also experienced an additional 12.2 hours per month of average time in light without pain at month six compared with placebo, with a p-value of 0.0010. Secondary endpoints showed statistically significant reductions in phototoxic reactions, improvements in light tolerance, and better patient-reported measures of disease severity and quality of life. Together, these findings support the potential for a treatment designed to address an underlying biological driver of EPP.
Disc reported that bitopertin was generally well tolerated, with dizziness the most commonly reported treatment-emergent adverse event in the treatment group, affecting 50% of participants. These events were generally mild or moderate and transient, and no treatment-emergent adverse events above Grade 3 were reported. The randomized, double-blind study enrolled 183 patients and evaluated once-daily oral treatment over six months.
Disc plans to resubmit its New Drug Application (NDA) to the U.S. Food and Drug Administration by the end of 2026, with an FDA decision anticipated by mid-2027. That timeline gives investors a clear series of upcoming milestones, although the earlier Complete Response Letter means regulatory execution remains central to the story.
Silence Therapeutics: Proposed $1.3B acquisition puts spotlight on RNA innovation
Silence Therapeutics (NASDAQ:SLN) entered into a definitive agreement to be acquired by Merck KGaA (OTC:MKGAF), not to be confused with Merck & Co. (NYSE:MRK), in an all-cash transaction valued at approximately $1.3 billion in equity value and $1.1 billion in enterprise value. Under the announced terms, shareholders will receive $20.00 per American Depositary Share (ADS), with each ADS representing three ordinary shares. The proposed price represents a 130% premium to Silence’s closing ADS price on October 16, 2026, and an 82% premium to its 30-day volume-weighted average price. The agreement gives shareholders a defined proposed cash consideration while highlighting the strategic value Merck sees in the company’s technology and drug pipeline.
A key asset is divesiran, an investigational small-interfering RNA (siRNA) therapy being developed for polycythemia vera, a rare blood cancer characterized by excessive red blood cell production. Divesiran is designed to silence TMPRSS6, a gene involved in regulating hepcidin and iron metabolism. By increasing hepcidin activity and limiting iron availability to the bone marrow, the treatment aims to reduce excessive red blood cell production. Silence has reported encouraging Phase 2 results and is positioning the candidate for further development. Merck’s resources could help advance the program, although future clinical success and commercial potential remain unproven.
The acquisition also expands Merck’s access to Silence’s proprietary mRNAi GOLD platform, which is designed to develop precision-engineered medicines that silence genes associated with disease. The broader pipeline includes programs addressing cardiovascular risk and other conditions with significant unmet medical needs. RNA-based therapies have become an important area of pharmaceutical investment because gene silencing offers potential approaches to biological targets that may be difficult to address with conventional drugs. Nevertheless, each candidate faces its own development, safety, regulatory, and commercial challenges.
The transaction is expected to close in the first quarter of 2027, subject to shareholder approval, court sanction, regulatory clearances, and other customary conditions.
NeOnc: Early patient improvement draws attention to investigational brain cancer treatment
NeOnc (NASDAQ:NTHI) reported early clinical improvement in a three-year-old boy with an aggressive brain tumor who is receiving investigational NEO100 through a compassionate-use program in Abu Dhabi. According to the child’s treating physicians, he is breathing independently without ventilator support and beginning to move his legs following initiation of treatment. The report has drawn attention to NEO100, a highly purified form of perillyl alcohol administered intranasally and designed to provide a non-invasive route for delivering treatment to the brain.
The circumstances surrounding the treatment are important. The child did not meet the eligibility criteria for NeOnc’s pediatric NEO100-03 clinical trial and is receiving treatment outside that study through the company’s Compassionate Care program. Access was coordinated among NeOnc, the treating physicians, and Abu Dhabi’s Department of Health. Compassionate-use programs can provide access to investigational treatments for patients with serious or life-threatening conditions who lack satisfactory alternatives, but they do not establish that a treatment is effective.
NeOnc is also advancing a broader clinical development program. The company announced regulatory authorizations in Abu Dhabi covering its NEO100-01, NEO100-02, and NEO100-03 protocols, spanning adult studies and a pediatric development pathway subject to further protocol review. In March 2025, it announced that the FDA had granted Rare Pediatric Disease Designation to NEO100 for pediatric-type diffuse high-grade gliomas. The company has also reported that its oral candidate NEO212 completed Phase 1 dose escalation and is advancing toward Phase 2 development. These milestones provide context for the company’s ambitions in central nervous system cancers, but designations and trial authorizations are not equivalent to marketing approval.
Alaunos Therapeutics: $350M financing supports new pulmonary hypertension strategy
Alaunos Therapeutics (NASDAQ:TCRT) completed its acquisition of Velaris Therapeutics alongside a private placement that generated approximately $350 million in gross proceeds. Announced on October 13, 2026, the transaction shifts the company’s focus toward a portfolio of investigational biologics for pulmonary arterial hypertension (PAH) and related forms of pulmonary hypertension. Management expects the financing to fund operations into 2029, providing resources to pursue multiple development milestones. In a capital-intensive industry, that runway is a meaningful component of the company’s investment story.
Velaris’s lead candidate, VLR199, is an activin type IIA receptor-Fc ligand trap designed to address signaling pathways involved in pulmonary vascular remodeling. The program aims to build on the clinically validated biology of activin-signaling inhibition while seeking improvements over existing therapeutic approaches. The pipeline also includes VLR247 and VLR258, designed to spare BMP-9 and BMP-10, ligands associated with certain hematologic adverse effects, and VLR360, an antibody targeting PDGF-B, a complementary pathway implicated in vascular remodeling.
The financing is intended to support a series of future catalysts rather than an immediate commercial launch. Alaunos expects clinical proof-of-concept data for VLR199 in 2028, alongside additional development milestones for VLR360 and initial regulatory filings for VLR247 and VLR258. That schedule gives the company time to test its scientific approach while reducing its near-term dependence on additional fundraising.
